1,3,3-trimethyl-N-(2-methylpropylidene)-5-[(2-methylpropylidene)amino]cycl ohexanemethylamine was tested in different aquatic assays to examine the short term toxicity.


The test substance was tested in the fish acute toxicity test according to the OECD Guideline 203 (July 1992) using Danio rerio (Hamilton-Buchanan) as test organism. The experiment was carried out as a static limit test, i.e. the test substance preparations were not exchanged for the whole exposure period of 96 hours. The results of this study show that the test substance does not have any impact on Danio rerio at the tested concentration of 100 mg/L (nominal). Hence the LC50value after 96 h of exposure was determined a >100 mg/L, based on nominal concentration. Based on the measured DOC value the LC50 value after 96 h was > 53.7 mg/L.


In the acute Daphnia toxicity test according to the OECD Guideline 202 (April 2004) Daphnia magna (Straus) was used as test organism. The experiment was carried out as a static test, i.e. the test substance preparations were not exchanged for the whole exposure period of 48 hours. The test was performed with test substance preparations of nominal 3.75 mg/L, 7.5 mg/L, 15 mg/L, 30 mg/L, 60 mg test substance/L M4 medium. The EC50value after 48 hours of exposure was 22.2 mg/L and the NOEC was 7.5 mg/L, based on nominal concentrations. Based on measured DOC values, the EC50value was 14.7 mg/L.


The toxicity to algae was tested in the alga growth inhibition test according to the OECD Guideline 201 (23 March 2006) using the single cell green algae Desmodesmus subspicatus as test organism. The experiment was carried out as a static test, i.e. the test substance preparations were not exchanged for the whole exposure period of 72 hours. Based on non-GLP pre-test results the main test was performed with test substance concentrations of nominal 12.5, 50.0 and 100 mg/L mg/L culture medium. During this study the DOC of the test substance concentrations of nominal 6.25, 25, and 100 mg/L culture medium and the control (culture medium) was determined with a TIC analyzer. The results of this study show that the test substance induced inhibition of algal growth after 72 hours of exposure. For the parameter growth rate the ErC50 value (72 h) was >100 mg/L and the LOEC and NOEC values were 25 mg/L and 12.5 mg/L, respectively, based on nominal concentrations. Based on measured DOC values (0 h) the ErC50 value was 73.6 mg/L and the NOEC was 7.6 mg/L.


The influence of 1,3,3-trimethyl-N-(2-methylpropylidene)-5-[(2-methylpropylidene)amino]cyclohexanemethylamine on the activity of activated sludge was evaluated by measuring the respiration rate under defined conditions according to OECD 209 and EU method C.11. The respiration rate (oxygen consumption) of an aerobic activated sludge fed with a standard amount of synthetic sewage was measured in the presence of various concentrations of the test item after an incubation period of 3 hours. Five concentrations ranging from 10.0 to 1000 mg/L were tested. The results of the study indicate that the test substance does not effect aquatic microorganisms up to a concentration of 32 mg/L. From 100 mg /L onwards the respiration rate of the activated sludge was inhibited. The EC50 was 304.2 mg/L for the overall respiration and 405.9 mg/L for the heterotrophic respiration (without nitrification).
