The test substance concentrations of the blank control and the treatment group were sampled and analyzed when freshly prepared and prior to renewal during the firstweek of the test. 2OmL test solution was taken respectively from the freshly prepared test solutions and the solutions prior to renewal at each group and filtrated with O.22pm cellulose nitrate membrane filters. In Week 2 and 3, the sampling was operationed twice a week, once after renewal, and the next before renewal.
The samples were stored in refrigerator (2-8'C) after pretreatment (see 2.2.7) if thesamples were not analyzed directly in 24h.

PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
Range-finding test
According to the information provided by the sponsor, test substance is insoluble in water ( <0.4µg/L), 2L solution of the test substance, with a nominal concentration of 10 mg/L, were prepared with Elendt M4 medium and stirred (via magnetic stirrer) in the dark for 24h with the vortex depth of 1/3 for the solution, and were filtrated with 0.45 µm nitrocellulose membrane to make a 100% saturated solution of the test substance in the test water (the first 100mL of the filItrate were discarded), which was used as the stock solution and the test solution of the highest treatment group, The test medium of the control group was treated in exactly the same way as the highest treatment group, including stirring and filtration. The test solutions of the other treatment groups were prepared by diluting the 100% saturated solution with Elendt M4 medium.
The pH of the stock solution measured during the test was 8.061 - 8.165, not exceeding 6 - 9, and needed not to be adjusted.

Limit test
2L solution of the test substance, with a nominal concentration of 10 mg/L, was prepared with Elendt M4 medium and stirred (via magnetic stirrer) in the dark for 24 hours with the vortex depth of 1/3 for the solution, and were filtrated with 0.45 µm nitrocellulose membrane to make a 100% saturated solution of the test substance in the test water (the first 100mL of the filtrate were discarded), which was used as the test solution of the teratment group. The test medium of the control group was treated in exactly the same way as the treatment group, including stirring and filtration.
The pH of the treatment group measured during the test was 8.208 - 8.339, not exceeding 6 - 9, and needed not to be adjusted.
