The short hatching of the earthworms before introduction in the test was carried out with a pH-value of 4.1. However this has no influence on the EC-value of the reference substance, the weight and the clitellum of the earthworms.

Validity criteria:

- The mortality in the controls did not exceed 10 % at the end of the test.

- The LG50 <14d) of the control substance chloracetamide was 25,9 mg/kg.

Additional information

All substances in this category share a similar structure with a molecular weight > 500 g/mol. Based on the high molecular weight and insolubility, the substances in this category will not be able to penetrate biological membranes. Thus, no systemic exposure is expected. This is confirmed by the environmental studies on soil microorganisms except arthropods for one substance in this category: CAS 1328-53-6. The environmental toxicity data can be used for read-across to all substances in this category.


In a guideline study according to OECD TG 207 on Eisenia foetida, the substance CAS 1328-53-6 was mixed with artificial soil, and the earthworms were kept on the soil/substance mixture for 14 days. The 14-d NOEC was determined to be ≥ 1000 mg/kg soil (BASF 2000, K2). The results indicate that the substance is not toxic to soil macroorganisms (arthropods excluded).


In addition to the experimental data, all substances in this category have a low potential for adsorption to soil and no bioaccumulative properties. Therefore, binding to sewage sludge is unlikely. Consequently, a transfer to the soil compartment is not expected. The lack of harmful effects to all three trophic levels of aquatic organisms at concentrations below the solubility in water indicates a low hazardous potential of the test substance towards soil organisms. Based on the available information, it is concluded that all substances in this category are not toxic to soil microorganisms except arthropods, with an LC50 > 1000 mg/kg soil dry weight.
