Acute toxicity

The acute oral median lethal dose (LD50) of benzidine-2,2'-disulphonic acid in Rodent - rat was found to be 3400 mg/kg of body weight. Acute oral toxicity of benzidine-2,2'-disulphonic acid to Rodent-rat by oral route indicates that benzidine-2,2'-disulphonic acid does not exhibits acute toxicity by the oral route.

 

Since the vapour pressure of this chemical is very low (0.000000000000000431 Pa), there is least likelihood of exposure via the inhalation route. Therefore, this end point was considered for waiver

 

An experimental LD50 value of > 12000 mg/kg bw in rabbit was obtained for the read-across chemical (CAS No 131-11-3). In the absence of test data for the target chemical (CAS No 117-61-3) it is assumed that given the 60% structural similarity of the analogue to the target, benzidine-2,2'-disulphonic acid is assumed to not exhibit acute toxicity by the dermal route

 

Irritation / corrosion

From the available data, it can be concluded that the chemical benzidine-2,2'-disulphonic acid is unlikely to exhibit irritating effect on the skin but is slightly irritating to the eyes of rabbit within the concentrations mentioned in the study.

 

Sensitisation

Benzidine-2,2’-disulphonic acid was estimated to be sensitizing to skin of human in accordance to the Danish EPA database that is considered to be an authoritative database

Benzidine-2,2’-disulphonic acid is unlikely to exhibit acute toxicity by the oral, dermal and inhalation route, shows irritation effect to eye, is not irritant to the skin, does not exhibit genotoxicity and is not a reproductive toxin within the dose levels mentioned in the end points.

 

In the absence of local effects following short-term or long-term exposure, no dose-response data are available and a quantitative dose descriptor is not derived. DNEL values for local exposure are therefore not calculated.
