Description of key information

Weight of evidence: Thiosemicarbazide by oral route to rats produced some changes in body weight and clinical signs of tested animals. The NOAEL was judged to be 2 mg/kg/day for males and females, NOEL of 0.4 mg/kg/day for males and NOEL of 2 mg/kg/bw for female in the tested concentration 0, 0.4, 2 and 10 mg/kg. For the tested concentration 0, 0.2,1 and 5 mg/kg, thiosemicarbazide produced  some changes in body and organ weight,thymus size and small clinical signs of tested animals. The NOAEL was judged to be 5 mg/kg/day for females,and 1mg/kg for males and the NOEL of 1 mg/kg/day for males and 5 mg/kg for females.

Conclusions:

Executive summary:

Thiosemicarbazide was tested by oral route to rats in the doses 0, 0.4, 2 and 10 mg/kg/bw. Dosing period was 42 days far males, for satellite females 42 days and for females 14 days before mating to day 4 of lactation. There were used 12 male animals per group (5 for recovery), 12 females animals and 5 satellite females. Recovery period was also applied and it was 14 days for males and satellite females. Animals were killed after treatment and examinated fro clinical signs, body weight and weight gain, histopathological findings. Offspring were sacrificied on day 4 after birth. According to the data from National Institute of Technology and Evaluation, high level (10 mg/kg/bw) of thiosemicarbazide by oral route to rats produced  decreases in bodyweight (males), convulsion and excessive response to stimuli in both sexes and one death of female animal. Therefore NOAEL was judged to be 2 mg/kg/day for males and females, NOEL of 0.4 mg/kg/day for males and NOEL of 2 mg/kg/bw for females.

Dosing perid: 14 days

Death occured in one animal of three in total in the doses of 10 mg/kg and thsid ead animal had cyanosis and pulmonary edema The decrease of locomotor activity, respiration rate, body waight in males. Males appeared to had incomplete eyelid opening. The value of ALT decreased and T-Cho increased in t males animals. Females had blakish region of mucosa in the glandular stomach. Increase of weight of kidney R and decrease of weight of Thymus Al,, R was seen in both sexes. These abnormalities are conected with doses of 10 mg/kg .In the doses of 2 mg/kg Decrease of thymus A,R and increase of Kidney R was evaluated . Because of these findings, the selected dose for the study were doses: 0,0.2, 1 and 5 mg/kg/day.

Conclusions:
Thiosemicarbazide by oral route to rats produced some changes in body weight,thymus size and small clinical signs of tested animals. The NOAEL was judged to be 5 mg/kg/day for females,and 1mg/kg for males and the NOEL of 1 mg/kg/day for males and 5 mg/kg for females.
Executive summary:

Thiosemicarbazide was tested by oral route to rats in the concentration 0, 0.2, 1 and 5 mg/kg/bw. Dosing period was 42 days far males, for satellite females 42 days and for females 14 days before mating to day 4 of lactation. There were used 12 male animals per group (5 for recovery), 12 females animals and 5 satellite females. Recovery period was also applied and it was 14 days for males and satellite females. Animals were killed after treatment and examinated for clinical signs, body weight, weight gain, histopathological findings, food consumption. Offspring were sacrificied on day 4 after birth. According to the data from National Institute of Technology and Evaluation, high level (5 mg/kg/bw) of thiosemicarbazide by oral route to rats causes athropy and decreases in absolute and relative weights of the thymus in males and no changes were observed in females in the same concentration of 5 mg/kg. Temporary decrease of food consumption and total cholesterol were observed in males given 5 mg/kg and no effects were seen in females.The NOAEL was judged to be 5 mg/kg/day for females,and 1mg/kg for males and the NOEL of 1 mg/kg/day for males and 5 mg/kg for females.

Weight of evidence: Thiosemicarbazide by oral route to rats produced one death of female, convulsion and excessive response to stimuli in both sexes and decrease of body weight in males. The NOAEL was judged to be 2 mg/kg/day for males and females, NOEL of 0.4 mg/kg/day for males and NOEL of 2 mg/kg/bw for female in the tested concentration 0, 0.4, 2 and 10 mg/kg. For the tested concentration 0, 0.2,1 and 5 mg/kg, thiosemicarbazide produced  some changes in body and organ weight,thymus size and small clinical signs of tested animals. Males show an athropy and decrease in weights of thymus and females does not show any abnormalitites. The NOAEL was judged to be 5 mg/kg/day for females,and 1mg/kg for males and the NOEL of 1 mg/kg/day for males and 5 mg/kg for females.

Justification for classification or non-classification

Although there is description of "a disorder being done to a nervous systems by a repeated doses", there is no data which endorses these description, and it cannot be classified according to CLP Regulation (EC) no. 1272/2008.due to insufficient data.
