Description of key information

For aquatic toxicity results on short-term toxicity to fish and invertebrates as well as results on toxicity to algae and microorganisms are available.

Fish:

In the 48-h acute toxicity study according to DIN 38 412, Leuciscus idus ('Goldorfe') the 48-h acute toxicity LC50 of 4-chlorobenzonitrile was determined to be 22 mg/L (Hooftmann et al., 1990). The 24-h LC50 and NOEC values, based on mortality, were 24 and 18 mg/L, respectively. Due to undissolved test material that could be found at the bottom of the test vessels at 18 mg/L and in addition at the water surface at 56 mg/L, mortality that occured at concentrations of 18 mg/L and higher may be attributed to the interaction of the undissolved test material with the test organisms.

 

The 48-h LC50 for fathead minnow, calculated in a QSAR approach (Zhao et al., 1993), was determined to be 14.4 mg/l. This value is lower than the calculation of the 96-h LC50 of 4-chlorobenzonitrile for fish of 61 mg/L using EPISUITE 3.12 (US-EPA 2004; reported in BUA Stoffbericht 265, 2006).

Invertebrates:

Due to the results of a test according to OECD 202, the EC50 (48 h) of 4-Chlorobenzonitrile for immobilisation was determined to be 16.1 mg/L (nominal) and 8.06 mg/L (actual) of test item. The corresponding NOEC (48 h) was 6.25 mg/L (nominal) and 2.02 mg/L (actual).

Algae:

Significant inhibitory effects for 4-Chlorobenzonitrile were observed at 50.0 and 100 mg/L (nominal) for growth rate and yield, respectively. Therefore the overall LOEC was determined to be 50.0 mg/L (nominal) and the overall NOEC was 25.0 mg/L (nominal) for growth rate and yield. The 72h-EC50 value for growth rate (ErC50) was 51.9 mg/L (nominal). The 72h-EC50 value for yield (EyC50) was 20.5 mg/L (nominal).

Microorganisms:

A toxicity threshold value of 4-chlorobenzonitrile of 50.6 mg/L for Pseudomonas putida was determined. The ‘Bewertungszahl' (Assessment figure or value for bacterio toxicity) is therefore 4.3 (Bewertung wassergefährdender Stoffe, 1979; Coenen, 1990).

The EC10 and EC50 of 4-chlorobenzonitrile as well as the NOEC and LOEC values were calculated by using probit analysis and the Williams-Test (BUA Stoffbericht 265, 2006), respectively, on the data presented by Coenen, 1990. The following results were presented:

- EC10 (18 h) = 52 mg/L

- EC50 (18 h) = 138 mg/L

- LOEC (18 h) = 52 mg/L

- NOEC (18 h) = 26 mg/L

In addition the EC50 of 4-chlorobenzonitrile was determined in a Microtox-test for the bioluminescent marine bacterium Vibrio fisheri.The EC50 was determined to be 8.7 mg/L.

The toxicity of 4-chlorobenzonitrile to the freshwater ciliate Tetrahymena pyriformis was determined in a 48-h TETRATOX-Test (results reported in (BUA Stoffbericht 265, 2006).

The 48-h EC50 was determined to be 137 mg/L (nominal concentration).
