Details on results:
MTDID 49922 is a liquid mixture at room temperature. The water solubility was determined for DiBrOE2RMA, DiBrORMA, and DiBrE4RMA over a period of 4 days (Time point 1, 2,3, and 4). The results of day 1 were not reported.

For DiBrOE2RMA, the average measured water solubilities are 0.219 mg/L (RSD% = 26%), 0.129 mg/L (RSD% = 25%) and 0.207 mg/L (RSD = 37%) for Time Points 2, 3, and 4, respectively. For DiBrORMA, the average measured water solubilities are 0.0558 mg/L (RSD% = 50%), 0.0193 mg/L (RSD% = 82%) and 0.0514 mg/L (RSD = 93%) for Time Points 2, 3, and 4, respectively. For DiBrE4RMA, the average measured water solubilities are 0.318 mg/L (RSD% = 6.5%), 0.254 mg/L (RSD% = 2.3%) and 0.305 mg/L (RSD = 8.7%) for Time Points 2, 3, and 4, respectively.

The inter-day average measured water solubility of the DiBrOE2RMA is 0.183 mg/L (RSD=29%, n=27), the average water solubility of the DiBrORMA is 0.0422 mg/L (RSD=47%, n=27), and the average water solubility of the DiBrE4RMA is 0.292 mg/L (RSD=12%, n=27) at 22.2°C.

The study discussed that the high RSDs of DiBrOE2MA and DiBrORMA may be due to the MTDID 49922 minor components (mass ratio < 10%), undissolved materials, and low water solubilities. An effort was taken to separate undissolved DiBrOE2MA, DiBrORMA and DiBrE4RMA from water by centrifugation. The maximum centrifugation speed capability of 14000 RPM was used to separate them from the solution for 90 minutes. Several RSDs of OE2MA and ORMA were still > 15%. No further step was taken.

Applicant's summary and conclusion

Conclusions:
MTDID 49922 is a liquid mixture at room temperature. Three primary components of MTDID 49922 were measured for water solubility. Under the conditions of this study, the average measured water solubility of the DiBrOE2RMA was 0.183 mg/L (RSD=29%), the average water solubility of the DiBrORMA was 0.0422 mg/L (RSD=47%), and the average water solubility of the DiBrE4RMA was 0.292 mg/L (RSD=12%) at 22.2°C.
Executive summary:

The water solubility of MTDID 49922 was determined at ambient temperature using OPPTS 830.7840 “Water Solubility: Column Elution Method; Shake Flask Method”.

Triplicate test vials were prepared for each of the 4 time points (1, 2, 3, and 4 incubation days). The MTDID 49922 test substance was added to test vessel and 1.5 mL Milli-Q™ water was added. The test vessels were wrapped with aluminum foil and then incubated in a shaker at 30°C ±1°C and at a setting of 100 revolutions per minute (RPM). At the end of each time point, vials were removed from the shaker and allowed to equilibrate at ambient temperature (22.2 ± 0.6 °C) over 24 hours. After equilibration, the samples were centrifuged at a setting of 14000 RPM for 90 minutes. triplicate aliquots of each sample were removed from the aqueous phase and diluted 4-fold with methanol. All samples were analyzed using liquid chromatography/mass spectrometry (LC/MS).

This is a guideline study but conducted under GLP. OPPTS 830.7840 is recommended for the water solubility determination of pure substances. Since MTDID 49922 is a mixture, it is not known what effect, if any, the different components will have on each individual component measured water solubility. Additionally, there are high RSDs and estimated measurement uncertainties for DiBrOE2MA and DiBrORMA. Therefore, it is considered reliable with restriction and a key study.
