Details on inoculum:
The activated sludge used for this study was washed by centrifugation and the supernatant liquid phase was decanted.
The solid material was re-suspended in test water and centrifuged again. This procedure was done three times. An aliquot of the final sludge suspension was weighed, dried and the ratio of wet sludge to its dry weight was determined.
Based on this ratio, calculated aliquots of washed sludge suspension were mixed with test water corresponding to 4 g dry material per litre and aerated with CO2 free air overnight.

- Glass bottles with screw-caps and Teflon-coated septa (125 mL nominal, 128 mL total volume)

Details on results:
The 10-day window started at day 2, when the degradation was 17.1%. At sampling at day 9 (within the 10-day window), the degradation was 66.8%. Therefore, the ready biodegradation criterion was passed.
The degree of biodegradation of the test item is 68.3% in 16 days. At the end of the test at day 28, the degradation was 56.4%.
In the toxicity control containing both, the test item and the reference item sodium benzoate, 70.0% biodegradation was noted within 14 days and 58.1% biodegradation after 28 days of incubation. Therefore the test item is not inhibitory.

Conclusions:
The biodegradation of Saccharomyces cerevisiae, lysate estimated with the OECD 310 guideline, is 68.3% in 16 days and it satisfies the 10-day window.
Executive summary:

The substance Saccharomyces cerevisiae, lysate was tested for its ready biodegradability according to the OECD guideline No. 310 (CO2 headspace) and in compliance with the GLP.

The Inoculum was activated sludge microorganisms supplied by a municipal sewage treatment plant receiving domestic waste water.

The CO2 evolution resulting from the aerobic biodegradation of the test item by the inoculum was determined by measuring the inorganic carbon in the test flasks corrected by the control flasks. The extent of biodegradation was expressed as a percentage of the theoretical maximum IC production, based on the quantity of test substance initially added (as organic carbon). As a reference item, sodium benzoate was tested simultaneously under the same conditions as the test item, and functioned as a procedure control.

The test consisted of four groups :   

FT : test item (20.082 mg carbon/L)  + activated sludge and test water,

  FB : inoculum control with activated sludge and test water,

  Fc : procedure control with the reference item, sodium benzoate (19.976 mg Carbon/L), activated sludge and test water

  Fi : toxicity control (20.188 mg carbon/L) with test item, reference item (sodium benzoate), activated sludge and test water

The reference item sodium benzoate was sufficiently degraded to 74.5% after 14 days of incubation, thus confirming the suitability of the aerobic activated sludge inoculum used.

In the toxicity control containing both, the test item and the reference item sodium benzoate, 70.0% biodegradation was noted within 14 days. Therefore it can be assumed that the test item is not inhibitory to the aerobic activated sludge microorganisms.

The biodegradation of the test item reached 68.3% in 16 days.

The 10-day window started at day 2, when the degradation was 17.1%. At day 9 (within the 10-day window), the degradation was 66.8%. Therefore, the ready biodegradation criterion was passed.

In conclusion, Saccharomyces cerevisiae, lysate is designated as readily biodegradable, fulfilling the 10-day window.

This biodegradation test is classified as acceptable, and satisfies the guideline requirements of the OECD 310.
