- ln this study AMINOL (A098) was found to inhibit the growth of the freshwater green alga Desmodesmus subspicatus at nominal concentrations <: 25 mg/L (biomass growth and growth rate). The EC50-values with 95 % confidence intervals for inhibition of biomass growth (EbC50) and specific growth rate (ErC50) after 72 h were 17.9 (15.2- 21.2) and 56.0 (47.7- 65.7) mg/L, respectively. Inhibition effects were found to be reversible up to the nominal concentration of 100 mg/L (all effect levels given as nominal concentrations).
- Executive summary:
The toxicity of AMINOL (A098) to the unicellular freshwater green alga Desmodesmus subspicatus was determined according to the principles of OECD-Guideline 201 and the EC Directive 92/69/EC, C.3., at DR.U.NOACK-LABORATORIUM FüR ANGEWANDTE BIOLOGIE in 31157 Sarstedt, Germany from July 8 to 11, 2003. The aim of the study was to assess the NOEC, LOEC and EC50 values of
growth rate and biomass production over a period of 72 h. AMINOL (batch R0075037) contained 99.6 % (w/w) 2-(1 ,3-benzodioxole-5-ylamino)ethanolhydrochloride. The study was conducted under static conditions with an initial cell density of nominally 104 cells/ml. On the basis of a preliminary test, live concentrations of nominal 6.25- 12.5- 25- 50- 100 mg/L were tested in a geometrical series with a dilution factor of 2. Three replicates were tested for each concentration level and six replicates for the control. Microscopic evaluation of the cells at the start and end of the incubation period revealed no morphological abnormalities. Water quality parameters like pHvalue, measured at 0 and 72 h, and room temperature, measured continuously, were determined to be within the acceptable limits. The concentrations of 2-(1 ,3-benzodioxole-5 - ylamino)ethanolhydrochloride were analysed at all concentration levels at test start and test end via HPLC. The mean values of recovery rates of the active ingredient at the beginning of the test were in the range of 91 to 100 %. At test end concentrations of the active ingredient were < 0.1 mg Aminol/L (limit of quantification, LOQ). All effect values are given based on the nominal concentrations. After 72 h algae were transferred from the nominal dosage levels 25 - 100 mg/L and control to fresh untreated medium and allowed to grow for further 3 d under test conditions. The test item effect was observed to be reversible at these concentration levels.
