Description of key information
The skin sensitization potential of target chemical Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) was assessedin various experimental studies which were conducted on guinea for target chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) andits structurally similar read across substancesAcid Yellow 23 (CAS No: 1934-21-0)and Sodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6). The predicted data usingQSAR toolboxhas also been compared with the experimental data.Based on the available data for the target and read across substances and applying the weight of evidence approach, it can be concluded that chemical Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) is unable to cause skin sensitization and thus can be considered as not sensitizing. Comparing the above annotations with the criteria of CLP regulation, it can be classified under the category “Not Classified”.

Skin sensitization:
Various studieshas been investigated for the test chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7)to observe the potential for skin sensitization to a greater or lesser extent. The studies are based on in vivo experiments in guinea pigs for target chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) and its structurally similar read across substancesAcid Yellow 23 (CAS No: 1934-21-0)and Sodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6).The predicted data using the OECD QSAR toolbox has also been compared with the experimental data and summarized as below;
The skin sensitization potential of Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7)was estimated using OECD QSAR toolbox version 3.3 with log Pow as the primary descriptor. The chemical Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7)was estimated to be not sensitizing to the skin of Pirbright White guinea pigs. Based on the estimated result Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) failed to induce skin sanitization effects and hence is considered to be not sensitizing to Pirbright White guinea pigs.
The European Commission (EC) - Scientific Committee on Cosmetic Products and Non-Food Products (SCCNFP) {Opinion concerning: Acid Yellow 23 (Colipa n° C29); European Commission (EC) - Scientific Committee on Cosmetic Products and Non-Food Products (SCCNFP) SCCNFP/0786/04; 23 April 2004} reported a Maximisation (Magnusson and Kligman) Test on albino Himalayan spotted guinea pigs for read across chemical Acid Yellow 23 (CAS No: 1934-21-0) in accordance with OECD Guideline No. 406.During the induction, a 5 % Acid Yellow 23 solution (in 1 % carboxymethylcellulose solution; prepared with bidistilled water) was applied in an emulsion of Freund´s Complete Adjuvant (FCA) for the intradermal induction. 1 week later, following treatment with sodium lauryl sulfate (SLS) the epidermal induction was conducted for 48 h under occlusion with a 50 % Acid Yellow 23 solution.Animals were challenged after 2 week by epidermal application of Acid Yellow 23 (25 % solution) under occlusive dressing. Cutaneous reactions were evaluated at 24 and 48 h after removal of the dressing. For challenge reading, all animals were depilated 3 h before examination to remove the discolouration.Since none of the control as well as test animals showed any signs of contact sensitization at after challenge treatment , the chemical Acid Yellow 23 (CAS No: 1934-21-0) was considered to be not sensitizing to the skin ofalbino Himalayan spotted guinea pigs.
The above results were further supported by the experimental study reported by European Commission (EC) - Scientific Committee on Cosmetology (SCC) {Environment and Quality of Life - Reports (Seventh Series), European Commission (EC) - Scientific Committee on Cosmetology (SCC), 1988} for read across chemical Sodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6) on guinea pigs.During the induction phase, guinea pigs were induced intradermally with injections of 0.1 mL of 0.5% aqueous solution of test chemical on 3 times per day on 5 consecutive days.After a rest period of 4 weeks, animals were challenged with the same concentration and then observed for allergic effects. The treated guinea pigs did not reveal any sensitization potential. Hence the chemicalSodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6) was considered to be not sensitizing to the skin of treated guinea pigs.
Thus on the basis of available data for thetarget chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7)and its structurally similar read across substancesAcid Yellow 23 (CAS No: 1934-21-0)and Sodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6),it can be concluded thatchemical Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) is unable to cause skin sensitization and considered as non-skin sensitizer. Comparing the above annotations with the criteria of CLP regulation, it can be classified under the category “Not Classified”.
