No interspecies differences are expected due to local effects in the respiratory tract. Therefore, as a general rule a AF of 1 for remaining interspecies differences provides sufficient protection.

Selection of the relevant starting point

Inhalation exposure is typically the most relevant route for assessing occupational hazard and risk in humans. In a subacute (4-week) inhalation study with aerosol exposure of Metalink U to rats a NOAEC of 1.2 mg/m3 was determined (Kopf, 2016). At higher concentrations (3.1 and 11.1 mg/m3) significantly increased lung weights in correlation with toxicologically relevant changes in bronchoalveolar lavage parameters (PMN, LYM, LDH, Protein, GGT) were observed. Histopathological observations of minimal to slight hypercellularity of the broncho-alveolar region in all rats at 3.1 mg/m3 and above were seen. Macroscopically, lung-associated lymph nodes were increased in size at 3.1 mg/m3 (only females) and 11.1 mg/m3. In summary, effects from repeated exposure of rats to Metalink U are limited to effects on the respiratory tract (portal-of-entry effects). No indications of systemic toxicity were found.

Therefore, a worker DNEL for long-term inhalation exposure - local effects have to be derived.

 

Derivation of the DNEL long-term inhalation exposure - local effects

Starting point: 1.2 mg/m3 (NOAEC from subacute inhalation study)

Correction of the starting point according to ECHA Guidance, Chapter R.8 (Nov. 2012), Figure R.8-2:

corrected NOAEC = 1.2 mg/m3 x (6 h/d : 8 h/d) x (6.7 m3 : 10 m3) = 0.603 mg/m3

According to ECHA Guidance, Chapter R.8 (Nov. 2012), Section R.8.4 a series of assessment factors (AF) were applied to the corrected NOAEC from the subacute rat inhalation study and are summarized in the table below:
