Test solutions

Vehicle:
no
Details on test solutions:
The stock solution (100 mg/L test item were weighed out) was prepared with dilution water one day prior to application.
The stock solution was shaken with 20 rpm for 24 hours (rotating shaker 3040, GFL). Undissolved particles were removed by membran filtration (0.45 pm, RC, Machery-Nagel).
Limit concentration: Saturated solution*
The tested saturated solution prepared with 100 mg test item/L was visually clear throughout exposure. Due to the low solubility of the test item the measured concentrations of 4-Hydroxybenzoic acid behenylester in the fresh (0 hours) and old media (48 hours) of the saturated* solution were
below the limit of quantification (LOQ = 4 µg/L).

*Saturated solution: The maximum dissolved concentration of the test item that can be achieved under the test conditions in the test medium, acc. to OECD Series, No. 23 (2000)6.

Test organisms

Test organisms (species):
Daphnia magna
Details on test organisms:
Test organism: Daphnia magna STRAUS (Clone 5)
Reason for the selection of the test organism: Daphnia magna belongs, in accordance with the study guideline, to the suitable Daphnia types and is
bred at the test facility.
Origin: Institut fur Wasser-, Boden- und Lufthygiene (WaBoLu)
Breeder: DR.U.NOACK-LABORATORIEN, Käthe-Paulus-Str. 1, D-31157 Sarstedt, Germany
Culture: In glass vessels (2 - 3 L capacity) with approximately 1.8 L culture medium, at 20 ± 2 °C, in an incubator, 16 h illumination, illumination strength max. 20 µE-m^-2s^-1.
Culture medium: Elendt M4, according to Elendt (1990), modified to a total hardness of 160 to 180 mg CaC03/L.
Feeding: At least 5 times per week ad libitum with a mix of unicellular green algae, e.g. Pseudokirchneriella subcapitata, Desmodesmus subspicatus and Chlorella vulgaris, with an algae cell density of > 106ceils/mL

The acute immobilization of the test item 4 -Hydroxybenzoic acid behenylester to Daphnia magna (STRAUS) was determined according to OECD 202 (2004). The study was conducted as a limit test with the saturated solution of the test item as limit concentration under static conditions over a period of 48 h. 20 test organisms were exposed to the limit concentration and control.

The saturated solution and the control were analysed via LC-MS/MS after 0 hours (new media) and 48 hours (ofd media).

Due to the low solubility of the test item the measured concentrations of 4-Hydroxybenzoic acid behenylester in the fresh (0 hours) and old media (48 hours) of the saturated solution were below the limit of quantification (LOQ = 4 µg/L). All effect values are given based on the saturated solution prepared with 100 mg 4 -Hydroxybenzoic acid behenylester/L. The water quality parameters (pH-value and dissolved oxygen concentration), measured at 0 and 48 hours, were determined to be within the acceptable limits. The validity criteria of the test guideline were fulfilled.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
At the saturated solution prepared with 100 mg 4-Hydroxybenzoic acid behenylester/L. The effective concentration of the substance in the solution was below the LOQ of 4ppb (4 µg/l). No effect on Daphnia magna was observed at this concentration. Therefore the substance is not classified.
