Details on results:
The analytic procedure as outlined in section 3.4 was found to be linear between 120 µg/ml and 10 µg/ml and its detection limit was found to be 5 µg/ml (see RCC NOTOX study 0917/C608).
Concentrations of test substance in buffer and n-octanol samples were calculated using the absorbance coefficient at 480 nm in Milli-Q

An overall partition coefficient of < 0.02 was calculated. Expressed as its logarithm (base 10), the mean partition coefficient is <-1.70; individual values fall within the range of ± 0.3 log units and therefore meet the quality criterion. Temperature of measurement was 20 ± 1°C.

Conclusions:
In the study performed according to OECD107, adopted May 12, 1981 the partition coefficient (n-octanol/water) for Guanidine Nitrate was determined to be < 0.02 (log Pow: < -1.70) at 20 ± 1°C.

From the preliminary test a partition coefficient of 0.07 was calculated (test substance concentrations of 5 microg/ml (detection limit) and 69 microg/ml were calculated for the n-octanol and buffer phase, respectively).

The analytical procedure as outlined in section 3.4 was found to be linear between 120 microg/ml and 10 microg/ml and its detection limit was found to be 5 microg/ml The test substance was determined to be stable for at least 4 hours in Milli-Q water (results not shown).

Concentrations of test substance in buffer and n-octanol samples were calculated using the absorbance coefficient at 480 nm in Milli-Q.

Temperature of measurement was 20 ± 1°C. pH values of the water phases are shown in Table 1; pH of the buffer solution was 7.4.
 In conclusion, the partition coefficient (n-octanol/water) for Guanidine Hydrochloride was determined to be < 0.02 (log pow: < -1.70) at 20 ± 1°C.

Additional information

In the study performed by RCC NOTOX (1988) the partition coefficient (n-octanol/water) for Guanidine Nitrate was determined to be <0.02 (log Pow: <-1.70) at 20 ± 1°C. The study was performed according to OECD107, adopted May 12, 1981. To enable functionalities within Chesar v.1.2, the partition coefficient (here being out of range with a value of logKow of -1.7) was set to the minimum value of the range advised by the implemented EUSES program, i.e. to >-1.

Additional supporting data from the read-across substance guanidine chloride were included to demonstrate the similar physico-chemical profile of both substances, used to substantiate the justification for read-across, outlined in chapter 13 of technical dossier.
