Short-term toxicity to aquatic invertebrates

Description of key information

To determine the toxicity of p-nonylphenol-formaldehyde-diethanolamine, Mannich base to aquatic invertebrates, a GLP-compliant aquatic invertebrates toxicity test was performed in accordance with OECD guideline 202 (Toxi-Coop Zrt, 2013). In this study, 20 daphnids per concentration (4 groups of five animals) were exposed to nominal test substance concentrations of 1.3, 2.5, 5.0, 10.0, and 20.0 mg/L and a blank control for 48 hours under static conditions. Analytical verification of nominal test concentrations was performed. As analytically measured test item concentrations deviated more than ± 20 % of the nominal over the test period of 48 hours, the exposure concentrations were calculated as the geometric mean of the concentrations measured at start and at the end of the experiment. Analytical concentrations were 1.5, 3.5, 6.9 and 14.2 mg/L (the lowest nominal concentration of 1.3 mg/L was below the analytical quantification limit (LOQ) at the end of the test and therefore excluded from the result evaluation). After 24 hours exposure no immobility was observed in the control and in the whole concentration range. After 48 hours exposure all daphnids were observed as immobile in the highest test concentration (14.2 mg/L). In the concentration of 6.9 mg/L 60% immobility was noticed at 48-h. Further immobility or any abnormal behaviour in the lower concentrations or in the control was not detected. Based on these findings, the 48-h EC50 is determined at 6.5 mg/L.
