Single samples for analysis were taken from all test concentrations and the control.

Sampling: Frequency At the beginning and at the end of three intervals of 48 hours (nominal days 2 and 4, 8 and 10, 14 and 16).

At the end of the refreshment period, the replicates were pooled at each concentration before sampling.
Additionally, reserve samples of 3.0 mL were taken from all test solutions for possible analysis.

Details on test solutions:
The batch of TRIETHYL PHOSPHONOACETATE tested was a colourless liquid with a purity of 99.3% (area) and the item was completely soluble in test medium at the concentrations tested. No correction was made for the purity/composition of the test substance.

Preparation of test solutions started with a stock solution of 100 mg/L (range-finding test) or the highest test concentration of 4.6 mg/L (final test). No special treatment other than a magnetic stirring period of approximately 15 minutes was needed to completely dissolve the test item in test medium. The lower test concentrations were prepared by subsequent dilutions of the stock or highest test concentration in test medium. All final test solutions were clear and colourless.

- Biomass loading rate: Twice daily an amount of 0.25 ml of a Chlorella sp. suspension . On weekend days an amount of 0.50 ml was added in one single feed. This daily ration corresponded to 0.2 mg C/Daphnia/day, which is the recommended value for daily feeding per daphnid in the reproduction test according to the OECD Guideline 211.

- Results used to determine the conditions for the definitive study: No significant mortality was observed in the control and the two lowest test concentrations, while all daphnids exposed to the highest test concentration of 10 mg/L died within 3 days of exposure. The first recordings of the presence of eggs in the brood pouch and the first offspring were similar to the control in the two lowest test concentrations. The total numbers of offspring produced were also comparable between the control and the two lowest test groups.

Conclusions:
Under the conditions of the present study TRIETHYL PHOSPHONOACETATE did not affect survival, growth or reproduction of Daphnia magna at an average measured concentration of 3.6 mg/L after 21 days of exposure (NOEC).
Executive summary:

This study was designed to determine the chronic toxicity of triethylphosphonoacetate to Daphnia magna, according to the OECD 211 guideline and under GLP conditions. Exposure was conducted under semi-static conditions for 21 days.

Parental daphnids were exposed to triethylphosphonoacetate concentrations of 0.16, 0.38, 0.93, 1.7 and 3.6 mg/L and to a control treatment. The number of living adults and offsprings were counted daily. At the end of the test period, a maximum of 10% adults died. The reproduction rates are not significantly different from that of the blank control. The 21-day NOECs on Daphnia magna were equal to 3.6 mg/L based on survival and to 3.6 mg/L based on reproduction; showing that triethylphosphonoacetate is not harmful to aquatic invertebrates with long-lasting effects.

Description of key information

The 21-day chronic toxicity of TEPA to Daphnia magna was studied under semi-static conditions according to OECD guideline 211.  Daphnids were exposed to control and test chemical at measured concentration of 0.16, 0.38, 0.93, 1.7 and 3.6 mg/L for 21 days.


The 21-day NOEC for reproduction was 3.6 mg/L (the highest dose tested).
