Additional information

Based on the presented results 'Reaction mass of benzyl 2-ethylhexyl adipate and bis(2-ethylhexyl) adipate and dibenzyl adipate' has to be scored as very toxic to aquatic organisms with long lasting effects (aquatic hazard category chronic 1). Data on aquatic effects of 'Reaction mass of benzyl 2-ethylhexyl adipate and bis(2-ethylhexyl) adipate and dibenzyl adipate' are available on three trophic levels (fish, daphnia and algae) for acute toxicity, and two trophic levels (fish, daphnia) for long-term toxicity.


 


Fish


A 96h-LL50 of >100 mg/L was obtained on Danio rerio in a semi static test system. The study was conducted in accordance with Council Regulation (EC) No 440/2008, Method C.1 'Acute toxicity for Fish' (2008) which is in most parts equivalent to the OECD Guideline for Testing of Chemicals No. 203 'Fish, Acute Toxicity Test' (1992). A Water Accommodated Fraction (WAF) was used to test effects at a limit lethal loading of 100 mg/L.


 


Daphnia


An acute toxicity study in Daphnia magna was performed over 48 h and the effect on mobility was determined to be 48h-EL50 >100 mg/L. The study was conducted in accordance with the Council Regulation (EC) No 440/2008, Method C.2 ‘Acute toxicity for Daphnia’ (2008) which is equivalent to OECD Guideline for Testing of Chemicals No. 202 'Daphnia sp., Acute Immobilisation Test'. A Water Accommodated Fraction (WAF) was used to test effects at a limit effective loading of 100 mg/L.


 


Algae


In a test, the fresh water alga Desmodesmus subspicatus was exposed to 'Reaction mass of benzyl 2-ethylhexyl adipate and bis(2-ethylhexyl) adipate and dibenzyl adipate' for 72 h. The study was conducted in accordance with Commission Regulation (EC) No 761/2009 amending Regulation No 440/2008, Method C.3 ‘Freshwater Alga and Cyanobacteria, Growth inhibition test’ (2009) which is equivalent to OECD Guideline for Testing of Chemicals No. 201 (2006). A Water Accommodated Fraction (WAF) was used to test effects at a limit effective loading of 100 mg/L. A 72h-EL50 of > 100 mg/L was received.


 


Microoganisms


The effect towards the growth of activated sludge was investigated according to ISO 8192 -1986 (E) having an EC50(3 h) of > 1000 mg/L. Therefore, an adverse effect towards microorganisms and thus the cleaning capacity of sewage treatment plants is not expected.


 


In addition, long-term toxicity on fish and daphnia was tested in recent studies (ECT, 2020).


 


Fish (long-term)


In a flow-through approach according OECD (2013). OECD Guideline for Testing of Chemicals, No. 210 "Fish, Early-life Stage Toxicity Test" adopted on 26th July 2013 several biological parameters were monitored, i.e. hatching success, number of larvae hatching each day, time to start of hatching and end of hatching, cumulative mortality (Post-Hatch Success (Survival)), number of deformed or abnormally appearing larvae, number of organisms exhibiting abnormal behaviour, length of the surviving fish and wet weight of the surviving fish over the course of 35 d. Dry weight was monitored but according to OECD Guideline for Testing of Chemicals, No. 210 "Fish, Early-life Stage Toxicity Test" wet weight is the preferred parameter.


 


For all relevant biological parameters, a NOEC, LOEC and where applicable EC50 and EC20 of ≥ 72.5 µg/L (geometric mean of the measured test item concentration) was determined. For the wet weight of fish no distinct correlation was observed regarding the concentration of the test item. Therefore, no reliable ECx values were calculated for the parameter fresh weight of fish larvae at the end of the exposure period. Nevertheless, the EC10 was determined to be 107 % of a filtrate at a loading rate of 10 mg test item/L based on nominal concentrations, equivalent to 70.4 µg test item/L based on measured concentrations.


 


In conclusion no effect on fish was determined.


 


Daphnia (long-term)


In a “Daphnia Magna, reproduction test" according OECD TG 211 the toxicity of the Reaction mass of benzyl 2-ethylhexyl adipate and bis(2-ethylhexyl) adipate and dibenzyl adipate on daphnia magna was assessed.


 


Over the course of 21 d no concentration-response relationship was observed for parental daphnid survival/immobility resulting in a NOEC = 26.6 µg/L and a LOEC >26.6 µg/L.


 


For the endpoint "reproduction based on surviving F0 daphnids" the following effect concentrations were observed: EC10 = 4.05 µg/L, EC20 = 10.45 µg/L, NOEC 4.34 µg/L and LOEC = 8.68 µg/L
