Method: 10008 mg test item was dissolved in 1000 mL of growth medium with magnetic stirring to provide a stock solution (10000 mg/L). The pH
of the stock solution was adjusted from 4.78 to 8.55 then the stock solution was diluted in growth medium to provide the test solutions.

A qualification test on the alga culture was made on 5 June 2008 to assure the quality of the test animals. ErC50 was 1.5 mg/L, which confirmed that the culture was usitable as a test organism according to OECD TG no. 201.

The EC-values were calculated by means of graphic interpolation. The NOEC-value is the highest tested concentration not statistically different from the controls (Mann-Whitney U-test p>0.05).

Conclusions:
A 72 hour algal growth inhibition study, test species Raphidocelis subcapitata (formerly known as Pseudokirchneriella subcapitata and Selenastrum capricornutum), was performed to determine the toxic effects of the test substance Polyol PX. The test was conducted according to OECD test guideline 201, and performed to GLP. Based on nominal exposure concentrations the 72 hour ErC50 (growth rate) was calculated to be 639 mg/L and the corresponding no-observed effect concentration (NOEC) was 250 mg/L.
Executive summary:

In a 72 hour acute toxicity study, the cultures of the green alga Raphidocelis subcapitata (NIVA CHL 1) were exposed to Polyol PX at nominal concentrations of 0 (control), 31,2, 62,5, 125, 250, 500, 1000 mg/L under static conditions in accordance with the guideline OECD 201 - Algae, Growth inhibition test. The NOEC and EC50 values based on growth rate were 250 and 639 mg/L, respectively. The % growth inhibition in the treated algal culture as compared to the control ranged from 0 to 90.


 


There were no compound related phytotoxic effects.


 


This toxicity study is classified as supplementary and satisfies the guideline requirements for Algae, Growth Inhibition - toxicity study.

- Source: MBM Sciencebridge GmbH (Albrecht-von-Haller-Institut für Pflanzenwissenschaften Experimentelle Phykologie und Sammlung von Algenkulturen (SAG)).
- Method of cultivation: The algae are kept as stock culture on solid agar at 2 - 8 °C. From the stock culture, a permanent culture was prepared. From an aliquot of the permanent culture, the pre-culture was prepared.

ACCLIMATION
For the pre-culture an aliquot of the permanent culture was brought into nutrient medium and incubated under continuous lighting for 4 days under test conditions (Lighting: within the specified range of 4440 – 8880 lux; 21.4 – 22.1 °C). The resulting culture grew exponentially. Before usage, the pre-culture was checked for the absence of cell aggregates and the cell number of the culture was determined.

Conclusions:
One valid experiment was performed.
The study was performed using 5 concentrations ranging from 10 to 100 mg/L (nominal).
Inhibition of algae growth was not observed at any of the tested concentrations. Algal growth in the test solutions was even slightly higher than in the control. Therefore. no statistical evaluation was performed and the biological results were reported as a range.
Executive summary:

In a 72 hour acute toxicity study, the cultures of the Chlorophyta Desmodesmus subspicatus (SAG Strain Number= 86.81) were exposed to Polyol PX at nominal concentrations of 10, 18, 32, 56, 100 mg/L under static conditions in accordance with OECD 201 resp. EU C.3. The NOEC and EC50 values based on growth rate and yiels were 100 and >100 mg/L, respectively. The % growth inhibition in the treated algal culture as compared to the control ranged from -18.25 to 0.14.

This toxicity study is classified as acceptable and satisfies the guideline requirements for an acute toxicity study with algae and cyanobacteria.
