A GLP static Daphnia magna acute toxicity test was conducted in accordance with US-EPA TG 72-2 equivalent to OECD 202: Daphnia sp., Acute Immobilisation Test at nominal concentrations of 9.3, 15.6, 25.9, 43.2, 72.0 and 120 mg a.i./L, using 2 replicates per concentration each containing 10 Daphnia.   The 48-h EC50 value was determined to be 20.3 mg/L

A GLP static Daphnia magna acute toxicity test was conducted in accordance with US-EPA TG 72-2 equivalent to OECD 202: Daphnia sp., Acute Immobilisation Test at nominal concentrations of 9.3, 15.6, 25.9, 43.2, 72.0 and 120 mg a.i./L, using 2 replicates per concentration each containing 10 Daphnia.

A primary stock solution was prepared by weighing 1.4005 gram (g) of Busan 11-M1 and transferring it to a 1.0-L volumetric flask. The flask was brought to volume with test water. Because of the limited aqueous solubility of Busan 11-M1, the stock solution was stirred for 24 hours to promote maximum dissolution. The remaining precipitate was allowed to settle out for 3.5 hours and the clear supernatant was siphoned off to clean containers. The stock solution was analysed for dissolved Busan 11-M1 concentration as barium. Test solutions were prepared by adding appropriate volumes of primary stock solution to 1000-mL beakers and bringing the final volume to 600 mL with dilution water.

Two 200-mL aliquots from each beaker of test solution were poured into replicate test containers and 200 mL was preserved for chemical analysis. A dilution water control was maintained concurrently with the test solutions. The test animals (10 per replicate) were incubated in 300 mL glass petri dishes containing 200 mL of test solution and covered with a glass plate.

The following endpoint were determined:

24-h EC50: 22.2 mg/L (95% CL 20.3 to 54.1)

48-h EC50: 20.3 mg/L (95% CL 11.4 to 22.4)

24-h NOEC : 7.94 mg/L

48-h NOEC : 7.94 mg/L

It can be concluded that Busan 11-M1 was toxic to Daphnia magna at mean measured concentrations >7.94 mg/L. The 48-h EC50 was determined to be 20.3 mg/L.
